Two parts to this one. They have use against other players, although their effectiveness will be limited. Just because someone has 32 points of bluff should not mean that basically anything they say will be received as fact as long as they drop a roll after it. As for DMs, that depends entirely upon the individual DM and situation. I personally would ask for them in response to some corresponding attempted player action (calming an angry NPC, or convincing someone to allow you entry to a normally inaccessible location), less likely to need them as part of the actual quest itself (mainly because there's nothing quite so frustrating as to have a planned quest derail because none of the involved players has more than three points in diplomacy and you're wanting a roll of 25 to accomplish the goal!).
